Each capsule contains 75 mg of pregabalin, 750 mcg of mecopolamine, and 150 mg of alpha-lipoic acid.
Patients with hypersensitivity to pregabalin or any component of this product. Angioedema and hypersensitivity reactions have occurred in patients receiving pregabalin therapy.
- Because pregabalin is excreted unchanged in the urine, has little metabolism in humans (less than 2% of the dose recovered in the urine as metabolites), and is not bound to plasma proteins, its pharmacokinetics are unlikely to be influenced by other factors through metabolic reactions or Protein binding offset.
- Lorazepam, oxycodone: Increased cognitive and overall performance
- Additive hypoglycemic effects may occur with concomitant use of antidiabetic agents and alpha-lipoic acid. Close monitoring of blood sugar is recommended when starting treatment with alpha-lipoic acid.
- Cisplatin: Alpha-lipoic acid antagonizes the action of cisplatin and may decrease the efficacy of cisplatin.
- Metal-containing products: Alpha-lipoic acid chemically reacts easily with metals (metal chelation) and, therefore, should not be administered with metal-containing products (eg iron preparations, magnesium preparations, dairy products due to their calcium content) as it may neutralize their effect.
- Pregastar Plus is a combination of pregabalin, mecobalamin and alpha lipoic acid. Pregabalin is a structural derivative of the inhibitory neurotransmitter, gamma-aminobutyric acid (GABA). Pregabalin does not bind directly to the GABAA, GABAB, or benzodiazepine receptors, and does not increase GABAA responses in cultured neurons. Pregabalin binds with great affinity to the alpha2-delta site (a helper subunit of voltage-gated calcium channels) in CNS tissues.
- Mecobalamin is a type of endogenous vitamin B12 coenzyme. It is well transported to neuronal organelles and promotes DNA and protein synthesis, axonal transport, regeneration of axons and myelination.
- Alpha-lipoic acid is a natural antioxidant that has a powerful antioxidant effect in the majority of body tissues. This action helps protect the neuron from reactive degradation products.
- It is primarily used in the treatment of diabetic peripheral neuropathy and is effective in relieving both somatic neuropathy and autonomic neuropathy in diabetes.
Dosage:
- Dosing should begin at 2 capsules daily in 2 divided doses and may be increased within 1 week based on efficacy and tolerability for a total of 4 capsules daily given in divided doses (2-3 times/day). Because pregabalin is eliminated primarily by renal excretion, the dose should be adjusted for patients with impaired renal function.
- Patients with Renal Impairment: In view of adverse dose-related reactions, and since pregabalin is mainly eliminated by renal excretion, the dose should be adjusted in patients with impaired renal function. Dosage adjustment in patients with renal impairment should be based on CLcr
- Pregastar Plus is given orally with or without food.
When stopping use of Pregastar Plus, the dose should be reduced gradually over at least 1 week
